چکیده

We conducted a survey of the phytosanitary status Prunus germplasm collection in Nikita Botanical Gardens, Yalta, Russia. The virome plants displaying virus-like symptoms was studied using Illumina MiSeq high-throughput sequencing. Reads related to prunus necrotic ringspot virus (PNRSV), prune dwarf (PDV), and ourmia-like 1 (OuLV1) were generated number samples. Near complete genomes two divergent PNRSV isolates, PDV isolate, contig partly covered OuLV1 genome assembled de novo metaSPAdes program. structure genomic RNA1, RNA2, RNA3 new ilarvirus isolates shown be typical PDV. This is first report characterization full-length from Russia, expanding information on their geographical distribution genetic diversity. An open reading frames (ORF)-based phylogeny all available GenBank divided each ORF into or three main clusters. A migrated one cluster another cluster, depending analyzed segment. different branching order may indicate reassortment evolutionary history some isolates.

Whole-Genome Characterization of Prunus necrotic ringspot virus Infecting Sweet Cherry in China

Prunus necrotic ringspot virus (PNRSV) causes yield loss in most cultivated stone fruits, including sweet cherry. Using a small RNA deep-sequencing approach combined with end-genome sequence cloning, we identified the complete genomes of all three PNRSV strands from PNRSV-infected sweet cherry trees and compared them with those of two previously reported isolates.

Expression of Prunus Necrotic Ringspot Virus Coat Protein in E. coli

Background and Amis: Serological assay is considered as one of the best choices for conducting large number of infection tests. Recombinant DNA technology has been used for expression of virus coat protein (CP) gene in prokaryotic bacterial cells such as Escherichia coli and the recombinant CP (rCP) is used as immunogen in antibody production. Heterologous CP protein expression and purification...

Adaptive covariation between the coat and movement proteins of prunus necrotic ringspot virus.

The relative functional and/or structural importance of different amino acid sites in a protein can be assessed by evaluating the selective constraints to which they have been subjected during the course of evolution. Here we explore such constraints at the linear and three-dimensional levels for the movement protein (MP) and coat protein (CP) encoded by RNA 3 of prunus necrotic ringspot ilarvi...

Vertical transmission of Prunus necrotic ringspot virus: hitch-hiking from gametes to seedling.

The aim of this work was to follow Prunus necrotic ringspot virus (PNRSV) infection in apricot reproductive tissues and transmission of the virus to the next generation. For this, an analysis of viral distribution in apricot reproductive organs was carried out at different developmental stages. PNRSV was detected in reproductive tissues during gametogenesis. The virus was always present in the ...

The Incidence and Genetic Diversity of Apple Mosaic Virus (ApMV) and Prune Dwarf Virus (PDV) in Prunus Species in Australia

Apple mosaic virus (ApMV) and prune dwarf virus (PDV) are amongst the most common viruses infecting Prunus species worldwide but their incidence and genetic diversity in Australia is not known. In a survey of 127 Prunus tree samples collected from five states in Australia, ApMV and PDV occurred in 4 (3%) and 13 (10%) of the trees respectively. High-throughput sequencing (HTS) of amplicons from ...
